Recommendations for the introduction of metagenomic high-throughput sequencing in clinical virology, part I: Wet lab procedure

نویسندگان

چکیده

Metagenomic high-throughput sequencing (mHTS) is a hypothesis-free, universal pathogen detection technique for determination of the DNA/RNA sequences in variety sample types and infectious syndromes. mHTS still its early stages translating into clinical application. To support development, implementation standardization procedures virus diagnostics, European Society Clinical Virology (ESCV) Network on Next-Generation Sequencing (ENNGS) has been established. The aim ENNGS to bring together professionals involved viral diagnostics share methodologies experiences, develop application recommendations. This manuscript aims provide practical recommendations wet lab necessary give development validation laboratory methods, including quality assurance, control assessment protocols.

The metagenomic approach and causality in virology

Nowadays, the metagenomic approach has been a very important tool in the discovery of new viruses in environmental and biological samples. Here we discuss how these discoveries may help to elucidate the etiology of diseases and the criteria necessary to establish a causal association between a virus and a disease.

Evaluation of a Pooled Strategy for High-Throughput Sequencing of Cosmid Clones from Metagenomic Libraries

High-throughput sequencing methods have been instrumental in the growing field of metagenomics, with technological improvements enabling greater throughput at decreased costs. Nonetheless, the economy of high-throughput sequencing cannot be fully leveraged in the subdiscipline of functional metagenomics. In this area of research, environmental DNA is typically cloned to generate large-insert li...

Tumour virology in the era of high-throughput genomics

With the advent of massively parallel sequencing, oncogenic viruses in tumours can now be detected in an unbiased and comprehensive manner. Additionally, new viruses or strains can be discovered based on sequence similarity with known viruses. Using this approach, the causative agent for Merkel cell carcinoma was identified. Subsequent studies using data from large collections of tumours have c...
